Using a Nix Spectro2 to differentiate milled flour of red lentil genotypes

Overview
Objectives

Test the accuracy of the Nix Spectro 2 on processed flour products (specifically lentil) to determine if it comparable to the HunterLab colorimeter which is the current industry standard.

Experiment Summary

We validated the use of the Nix Spectro 2 to assess food color by observing the results obtained with the Nix to those generated by the HunterLab (an industry standard). The a* and b* Nix scores were comparable to those obtained with the HunterLab (r2 = 0.9-0.95). L* scores had lower correlation values: 0.61 and 0.58 when comparing HunterLab (C) with Nix (D50) and Nix (C), respectively. Lower L* correlations could be a result of the influence of direct light exposure during HunterLab scans.

Experimental Design
  • Field trials were arranged in a Randomized Complete Block Design (RCBD) with seed for this experiment being taken from a subset of the site-years of the full experiment. This resulted in four (Sutherland2022, Rosthern2022) and three (Osimon2022) repetitions of each lentil genotype. See the original experiment for more details: Lentil Red cotyledon intensity: Lentil germplasm over 8 site-years in 2 macroenvironments from 2022-2023 using BELT.

  • There were 3 technical replications per colorimeter done on every sample when measuring the colour values. All samples were measured with each of 1) HunterLab Mini XE Plus, 2) Nix Spectro 2 with illuminant C, 3) Nix Spectro 2 with illuminant D50.

  • Seeds from all replications within a site-year where combined for each genotype. Whole seeds were collected at maturity and dried at 45°C until a moisture content of <13% was reached. Clean, dried seeds were dehulled using a Satake TM05C huller and finely ground using a Udy cyclone sample mill through a #60 mesh size (0.25 mm).

Pressed Flour CIE L*, a*, b* scores (Excel, TSV)

This dataset consists of the CIE L*, a*, and b* scores for dry pressed flour samples. Flour samples were obtained from seeds grown in the Sutherland2022, Rosthern2022 and Osimo2022 site-years of this linked experiment: Lentil Red cotyledon intensity: Lentil germplasm over 8 site-years in 2 macroenvironments from 2022-2023 using BELT. Samples were measured with the HunterLab Mini XE Plus and the Nix Spectro 2 with illuminants D50 and C (referred to as "HunterLab", "Nix C" and "Nix D50" respectively). All samples were measured 3 times (technical replicates) and there were 4 biological/field replicates for Sutherland2022/Rosthern2022 samples and 3 biological/field replicates for Osimo2022. The field plot IDs between this file and those on the linked experiment match.
